Antioxidant and Collagen Stimulating Activities in Human Gingival Fibroblast Cell Culture of Plant Extracts

Abstract

Introduction: The aim of the study was to screen the antioxidant and collagen stimulating activities in HGF cell culture from aqueous and 95 % ethanol extracts of Centella asiatica and aqueous extract of Ananas comosus. Materials and Method : These study were in vitro test. Total phenolic content, antioxidation activities by DPPH, FRAP assay, cell proliferation of HGF by MTT assay and stimulating activity of collagen from HGF cell culture media were investigated.The hydroxyproline assay was used for analysis of collagen. Results: The aqueous extracts of Centella asiatica exhibited the highest total phenolic compound content as 88.85±1.17 µg GAE/g while 95 % ethanol extract of Centella asiatica exhibited the highest antioxidant activity by DPPH assay and FRAP assay as IC50 207.69±6.00 µg Vit C/g and FRAP value as 0.19±0.01 mM, respectively. For collagen stimulating activity in HGF cell culture at 500 µg/ml of 95 % ethanol and aqueous extract of Centella asiatica and aqueous extract of Ananas comosus showed the collagen content as 521.61, 435.19 and 344.14  µg/ml, respective. At 500 µg/ml of  all extracts showed the highest stimulating activity of cell proliferation in HGF. At 1000 ug/ml 95 % ethanol extracts of Centella asiatica appeared cross cell type-specific cytotoxicity. Conclusion : From all results, we found that all extracts showed the high antioxidant activity and collagen stimulating activity in HGF cell culture, therefore these extractes could be applicable as drug development for treatment of mouth ulcers.
